ACE is present in the brain and might play a modulatory role in multiple steps of the pathophysiological processes underlying neurodegenerative disorders. We report the clinical, neuroradiological, and neurochemical findings in a mildly affected patient with an ACE gene (ACE) deletion. Using different in vitro and in vivo experimental models for the study of the role of ACE and of the bradykinin-B(1) receptor (B(1)R) in the brain, we found evidence for a role of ACE in multiple steps of the pathogenesis of neurodegenerative disorders. Our findings suggest that the absence of ACE activity in the brain may produce an imbalance between amyloid-beta (Abeta) production and neurotoxic Abeta degradation, a phenomenon that is particularly evident in patients with an ACE deletion.
